The starting point is a detailed discussion of your needs and goals so we best understand which antibody services will be right for you. We will ask questions about the chemistry and biology of your antigen target. This information often reveals barriers or problems that require different or alternative approaches. The project plan then evolves; setting expectations, describing timelines, explaining cost options, and discussing possible solutions to anticipated problems. This is where our experience with similar targets comes in to play.
Green Mountain Antibodies can support you in developing a monoclonal and polyclonal antibody against your target. Both programs can be managed side by side, allowing your reagents to be used concurrently. The quantity of antibody required and final application will allow us to guide whether a mouse monoclonal or rabbit polyclonal antibody will be best for you. For anti-idiotypic programs, the goal is often to develop both a monoclonal antibody for PK assays and a polyclonal antibody for ADA assays.
If you do not have purified antigen, we can offer suggestions for obtaining recombinant protein or help with peptide selection, synthesis and conjugation. We’ll offer our opinions on the best way forward. Our knowledge of protein chemistry, immunochemistry, and assay development supports decision making throughout the process.
You might have a constrained budget or other limitations. You may have knowledge of previous antibody development efforts or information in the literature suggesting helpful approaches. Antibody Characterization Services
Green Mountain Antibodies can characterize the antibody to assure optimal selection and performance, as well as confirm lot to lot reproducibility of your selected antibody. Characterization services include:
- Relative affinity, matched pair confirmation, binning and koff rates via Gator screening
- Dynamic Light Scattering, SDS-PAGE, SEC purity reading
- Antibody DNA Sequencing
- ELISA confirmation screening
- Assay development services
Antibody Production and Purification Services
Our hybridoma development process includes multiple rounds of subcloning to assure your line is monoclonal and stable. The process allows us to select clones with higher antibody production levels.
After investing time and resources making a cell line that produces a monoclonal antibody, you will realize substantial savings (cost/mg) over purchasing the antibody from a commercial source. This also removes concerns of continuity of supply and inventory availability. The best antibody for your application will be available when you need it. GMAb produces antibody via fixed volume tissue culture vessels or bench-top bioreactors into the gram scale. Monoclonal antibody is then purified by Protein G and extensively characterized. Chemical labeling of the purified antibody with biotin, fluorescent dyes or proteins is available.
