Antithrombin Summary

Antithrombin, known as antithrombin III or ATIII, is a plasma protease inhibitor and member of the serpin gene family. ATIII inactivates several enzymes of the blood coagulation system. Rates of ATIII-mediated inhibition of thrombin, factor Xa and factor IXa are accelerated by up to several 1000-fold by heparin.

The tendency to form blood clots in the veins is known as thrombosis, a characteristic of a blood disorder known as antithrombin deficiency. Thrombophilia is the inherited tendency to form blood clots. Antithrombin in the blood limits coagulation, or the blood’s ability to clot.

Green Mountain Antibodies internally developed a mouse monoclonal antibody against antithrombin using human ATIII as the immunogen. GMA-800, also known as BDI205, binds human and baboon antithrombin by ELISA. The antibody does not cross-react with bovine, canine, or rabbit ATIII. It is an IgG1 isotype and does inhibit antithrombin activity in chromogenic thrombin assays.

Applications

Working Concentration: Approximately 1-5 µg/ml. Researcher should titer antibody in specific assay.

ELISA: Binds human and baboon antithrombin.

Inhibition: Inhibits ATIII activity in chromogenic thrombin assays.

Product Formulation: Lyophilized from a ≥1 mg/ml solution in 20 mM NaH2PO4 0.15 M NaCl, 1.0% (w/v) mannitol, pH 7.4. Concentration determined by absorbance measurement at 280 nm and using an extinction coefficient of 1.4 (ε0.1%). Reconstitute with deionized water.

Storage: Store lyophilized or reconstituted and aliquoted material at -20°C for prolonged periods. Avoid freeze-thaw cycles. Alternatively, add 0.02% (w/v) sodium azide to reconstituted solution and store at 4°C.
